Taq DNA Polymerase is a highly processive 5-3 DNA polymerase that lacks 3-5 exonuclease activity. It is a single polypeptide chain with a molecular weight of approximately 95 kDa.Taq DNA Polymerase was originally isolated from the thermophilic eubacterium Thermus aquaticus BM a strain lacking Taq I restriction endonuclease. The enzyme was cloned in E.coli.

The HotStart Taq DNA Polymerase is a chemically modified Taq DNA Polymerase, whose enzyme activities can only be activated after 3-5 minutes of incubation at 94o C. The HotStart Taq Polymerase uses amplification conditions for regular Taq DNA Polymerase, except no polymerase activity will be present before the onset of thermal cycling. This prevents nonspecific DNA amplification and primer dimer formation. The amplified products, up to 6Kb in length, contain single 3’-overhanging-A ends. This allows for TA cloning methods, if the amplified fragments need to be cloned.

RNA can be selectively synthesized from either strand of the insert DNA with different polymerases. Homogeneously labeled single-stranded RNA can be generated with this system. Transcripts can be non-radioactively labeled with biotin or DIG-11-UTP or radioactively labeled to high specific activity with [alpha-32P] or [alpha-35S]-labeled nucleotides.Synthesis of hybridization probes T7 RNA polymerase allows highly efficient production of homogeneously labeled RNA. This labeled RNA may be used as hybridization probes in Southern northern and dot blots as well as in situ hybridizations.Suitable labelsTranscripts can be nonradioactively labeled with biotin-16-UTP or DIG-11-UTP. Pwo DNA Polymerase is a highly processive thermostable 5AE-3AE DNA polymerase and possesses a 3AE-5AE exonuclease activity also known as proofreading activity. The enzyme has no detectable 5AE-3AE exonuclease activity. Pwo DNA Polymerase exhibits increased thermal stability with a half-life of more than two hours at 100 C compared to Taq DNA Polymerase half-life of less than 5 minutes at this temperature. Pwo DNA Polymerase-generated PCR products are blunt-ended and can therefore be used directly for blunt-end ligation without any pretreatment of the ends.

AS ONE PR Polymerase is a thermostable enzyme with PR ability, which can be used in primer extension reactions and other molecular biology applications. PR Polymerase exhibits both 5 prime to 3 prime DNA polymerase activity and 3 prime to 5 prime PR exonuclease activity. It is recommended for applications, which require extremely high fidelity or blunt ending. Optimal reaction conditions are achieved by using the 10x Ammonium buffer containing MgCl2 provided with the enzyme. 25 mM MgCl2 is also included separately, in case a higher MgCl2 concentration is required for a specific reaction.
